Animals were immunized with human kappa myeloma protein. Spleen cells were fused with P3X63Ag8.653 myeloma cells. Tested and found negative for ectromelia virus (mousepox). A feeder layer of mouse peritoneal macrophages or X-irradiated STO cells (see ATCC 56-X) is necessary for successful initiation of a culture from a thawed ampule. |

| Subculturing |
Medium Renewal: Every 3 to 4 days Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml. |
