Restriction digests of the clone give the following sizes (kb): HindIII--6.2; EcoRI--6.2; HindIII/EcoRI--5.0, 1.2; HpaI--6.2; BamHI--6.2. The depositor notes that the fibrinogen clones (ATCC 59706-59710) are suitable for applications in gene mapping or involving nucleic acid hybridization, but they are not suitable starting material for expression in mammalian cells. The clones do not encode the signal peptide necessary for assembly and secretion and one is known to lack a single nucleotide in the coding region. Gamma genomic DNA sequence is available from GenBank and includes the following numbers: J00134, J00135, M10014, X00086. Expresses intact fibrinogen gamma chain which cross-reacts with antisera and retains the ability to clump cells of Staphylococcus aureus Newman D(2)C. The polymorphism is at a KpnI site; the SacI digest is used to decrease the fragment sizes to detectable lengths. The ability of this probe to detect the KpnI polymorphism was verified by J.
