The insert contains the following restriction sites (nucleotide positions according to the published sequence): BglII--911; XbaI--1029. Restriction digests of the clone give the following sizes (kb): EcoRI--4.3; HindIII--4.3; PstI--2.7, 1.5; consistent with deposited DNA. This clone includes the final nucleotide positions 15-1451. The single large open reading frame extends from 80 to 1031. There are other in frame AUG codons at positions 309, 312, 377, 428, and 688. The insert is oriented such that linearizing with EcoRI and transcription with SP6 RNA polymerase gives the positive polarity (sense) transcript whereas linearizing with HindIII and transcription with T7 RNA polymerase gives the antisense strand. |
