Restriction digests of the clone give the following sizes (kb): EcoRI--3.0, 1.3, 1.15; PvuII--2.4, 1.1, 0.95, 0.75, 0.35; SacI--2.7 (doublet); SacI/BglI--1.4, 1.15, 0.9, 0.85, 0.65, 0.4; SmaI--3.7, 1.2, 0.75; SmaI/SacI--2.7, 1.2, 0.9, 0.75. The insert contains the following restriction sites (nt from the 5' end): PstI--106, 752, 1741, 1905, 1998, 2208; PvuII--162, 1077, 2144; SmaI--863, 1569; NotI--953; EcoRI--1099. Contains a complete open reading frame of 444 nucleotides, with 5' and 3' flanking sequences. A 0.7 kb SmaI fragment containing the coding region (nt 863 -1569) detects a 2.3 kb mRNA in human erythroleukemia K562 cells. A 1.2 kb SmaI/SacI fragment from the 3' region (nt 1570 - 2738) detects a 1.5 kb mRNA in SV40-transformed human fibroblasts. The intact insert can be separated from the vector by a SacI/SmaI double digest. |
