Restriction digests of the clone give the following sizes (kb): EcoRI--4.8, 2.3; HindIII--5.5, 2.7; PstI--4.8, 2.1, 1.1; BamHI--5.6, 2.2; AccI--3.7, 2.9, 1.5. The insert contains SacI (2) and BamHI (1) sites. The depositor indicates a similar clone has been sequenced in another lab. This probe spans a large part of the region homologus to v-ros. Allele A1 was seen in 1/39 cell lines; A2 in 37/39 cell lines, and both present in 1/39. PstI digestion of genomic DNA gives 3 bands of 5.3, 3.2, 1.3 kb in 19/19 cell lines. An additional band of 2.65 kb was detected in 1 of the 19. Enzyme(s) not detecting polymorphism: BamHI, EcoRV, SstI. The genomic fragment sizes detected are (kb): BamHI--11, 3.15; EcoRV--12; SstI--greater than 30, 16, 1.5. |
