The insert contains the following sites (in bp from an end): Sau3A (20), HaeIII(52), PstI (85), HinfI (181), HaeIII (227). Restriction digests of the clone give the following sizes (kb): EcoRI--3.2, 0.35; BamHI--3.6; HindIII--3.6; KpnI--3.6; HincII--3.6. The first half of the dimer is 85% identical to the sequence of the 1' monomer; the second half is 80% identical to the 2' monomer. The insert is a dimer of the 171 bp consensus sequence of alphoid DNA from the subfamily of an alpha-satellite DNA made up of EcoRI dimers or multiples thereof. |
