The insert includes the following restriction sites (in nucleotide positions from the 5' end of the published sequence): BamHI--748, 1083, 1177; PstI--1586, 3524, 3999, 4060; XbaI--2359. Restriction digests of the clone give the following sizes (kb): BamHI--6.4, 0.8; EcoRI--4.6, 2.7; HindIII--3.2, 2.2, 1.3, 0.7; PstI--3.1, 1.9, 1.5, 0.5; XbaI--5.2, 2.4. The insert extends from nucleotide 71 to 4661 of the published sequence, and encodes the complete reading frame (109 - 1495), plus 5' and 3' untranslated regions. This detects a mRNA of about 5.1 kb in normal and acute-phase rat liver. |
