Restriction digests of the clone give the following sizes (kb): XhoI/HindIII--3.0 (doublet), 1.3; KpnI--5.5, 1.0, 0.8; PstI--3.1, 2.4, 1.6, 0.23; PvuII--2.4, 1.8 (doublet), 0.43 (doublet), 0.32, 0.2; SmaI--3.3, 2.8, 0.7, 0.3, 0.17; XhoI--7.3; XhoI/SalI--4.3, 3.0. In southern blots, the probe detects other cross-reactive bands after hybridization at high stringency. The 3' end of the insert corresponds to nt 1073 (in the POU homeodomain) of the GenBank record. It includes sequence encoding the POU specific domain. Transcription from the T7 promoter gives the sense strand. |
