Restriction digests of the clone give the following sizes (kb): BamHI--4.9, 2.9; BstEII--8.0; SstI--7.2, 0.5; NotI--8.0; EcoRI--5.2, 2.6. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain), with mutations K38T, L39E, replace the corresponding region of factor IX. |
