The insert contains internal PstI, HindIII, BamHI BglII, XbaI, and NcoI sites. Restriction digests of the clone give the following sizes (kb): BamHI--2.8, 0.67, 0.48; SacI--4.0; XbaI--3.4, 0.69; SphI/EcoRI--2.45, 1.5; BglII--4.0; PstI--3.1, 0.94. The SphI site in the vector is 432 bp from the SalI cloning site. The SphI/EcoRI fragment contains about 500 bp of non-insert DNA (some from pSP64 and some from pcD). An XhoI fragment from MoIl3 (in a pcD vector) was cloned into the SalI site of pSP64. This fragment contains some of the SV40 promoter and polyadenylation sequences from the pcD vector. |
